Azure II, which is a blue-colored powdered stain, is a combination of an equal amount of Azure B and Methylene Blue. It has a molecular formula of C16H18N3SยทC15H16N3Sยท2Cl and also known as Azur II. This dye is commonly used in hematologic staining, particularly in conjunction with Eosin Y. The purple cell nucleus stain is generated by the molecular interaction of the Azure B-DNA complex and Eosin Y, with the intensity of the stain dependent on the concentration of the Azure II, and the ratio of the blue to the eosin color. Other variables like the pH of solutions, buffer solution, fixation procedure and staining time can influence the end staining outcomes.
Applications
- Widely employed in the Richardson stock solution to stain lysosomal glycogen.
- Used in Periodic Acid- Schiff (PAS) staining methods of glycogen, nuclei and cytoplasm.
- Azure II complexes can be used to stain nuclei, goblet cells, striated muscle, endothelial cells, smooth muscle, cytoplasm, lipid droplets, red blood cells, glycogen, collagen, cartilage, mucus granules, elastin and also leukocytes.
Key features
- Optimized for Hematologic Staining: Used with Eosin Y to obtain specific visual results in the cell population.
- Replicable and Stable Performance: Gives consistent and reliable staining values between samples.
- Wide Staining Capability: Useful in nuclear as well as cytoplasmic components.
- Water-Soluble Powder: Simple to use and manipulate in a laboratory setting.
- Multiplexable with Various Staining Protocols: Fits PAS procedures and Richardson staining, among other histological procedures.
