Azure ll is a blue colored powder which is a mixture of Azure B and Methylene Blue in equal amounts. Its chemical formula is C16H18N3S · C15H16N3S ·2 Cl. Azure ll is also known as 3-Chloro-6-hydroxy-2-Pyridinecarboxylic acid, Azur II. It is used for hematologic staining with added Eosin Y stain. The usual color of nuclei (purple) is achieved as a result of molecular interactions between the Eosin Y stain and the Azure B-DNA complex. Staining intensity depends on the amount of Azure II and Azure B/Eosin Y ratio. Several factors may contribute to the results of staining, such as pH solution and buffer solutions, buffer substances, fixation and duration of the staining procedure.

Application :
- Azure II has been used in the Richardson′s stock solution for the staining of lysosomal glycogen.
- It has also been used in the periodic acid schiff staining procedure for the staining of glycogen, nuclei and cytoplasm.
- Azure II complex is used for the staining of nuclei, goblet cells, striated muscle, endothelial cells, smooth muscle, cytoplasm, lipid droplets, red blood cells, glycogen, collagen, cartilage, mucus granules, elastin and leukocytes.
