KOH used in histology:A small piece of tissue is placed on a glass slide and a drop of 10% KOH is added. The slide is then covered with a coverslip and placed in a warm water bath for 3-5 minutes. This allows the KOH to dissolve the keratin and clear the cellular material. After the incubation period, the coverslip is removed and the slide is rinsed with water. The slide is then stained with a suitable stain, such as lactophenol cotton blue or periodic acid-Schiff (PAS).
