Methylene Blue Alkaline Loffler’s Solution is a staining solution that includes methylene blue dye in an alkaline environment. It is also referred to as Methylene Blue solution, Ehrlich’s reagent III, Loeffler’s Methylene Blue. The chemical formula of this solution is C16H18ClN3S. The solution is commonly made by dissolving methylene blue in a potassium hydroxide (KOH) or sodium hydroxide (NaOH) solution. Methylene Blue Loeffler reagent is utilized for staining acidoresistant bacteria based on Ziehl-Neelsen, utilizing the dye’s ability to stain nearly all the bacterial cell components in a slightly alkaline medium (Methylene Blue Loeffler reagent has a pH value of 8-8.5).
