Cotton Blue is a bluish black powder. Its chemical formula is C37H27N3Na2O9S3. It is soluble in water and slightly soluble in ethanol. Cotton blue is also known as Methyl blue. Cotton Blue Stain is formulated with lactophenol, which serves as the mounting fluid and the dye, cotton blue. An aniline dye called cotton blue stains the chitin in the cell walls of fungi, giving the fungus preparation color and highlighting and contrasting the structures. It is used as a stain in histology.

Methyl Blue is a histology dye commonly used to stain collagen blue in tissue sections. The compound has been determined useful as a vital dye for interocular observations. Methyl Blue has been incorporated in Sabouraud agar for the identification of Candida albicans strains. The compound has also been incorporated in other histology staining procedures such as Mallory’s connective tissue stain.
