Biebrich Scarlet is a red to brown colored powder. Biebrich scarlet is an azo organosulfur dye. Its chemical formula is C22H14N4O7S2Na2. Biebrich Scarlet is also known as C.I. 26905; Acid Red 66; Ponceau BS; Biebrich scarlet WS; Biebrich scarlet sodium salt. Biebrich scarlet, is an organic sodium salt having 2-[(2-hydroxynaphthalen-1-yl)diazenyl]-5-[(4-sulfonatophenyl)diazenyl]benzene-1-sulfonate as the counterion. It can be used as a plasma stain in Masson’s trichrome. One of the properties of Biebrich Scarlet dyes is that they do not change color in wide pH value ranges, which enables alkaline solutions of this dye to be used for selective staining of extremely basic proteins. The stain, Biebrich scarlet, is photo fixing and is especially useful for identifying eosinophils by binding to their cytoplasmic granules in tissue samples from 6 to 60 microns in thickness. It also stains granules of macrophage and granular lymphocytes.
